联苯代谢对微生物的生长胁迫及分裂抑制 总被引:1,自引:0,他引:1
以联苯/多氯联苯降解菌株R04(Rhodococcus sp.R04)和几种模式微生物为研究对象,利用高效液相色谱、荧光显微镜、扫描电子显微镜等分析微生物在联苯及其代谢物培养条件下细胞分裂和形态的变化.结果表明联苯及其代谢产物对红球菌R04和几种模式微生物细胞的分裂有抑制作用,并对部分微生物形态有影响.与前体-联苯及其代谢产物2-羟基-6-酮基-6-苯基-2,4-己二烯酸相比,2,3-二羟基联苯对G+、G-细菌,或是酵母细胞分裂都有较强的抑制和形态的改变.2,3-二羟基联苯导致R.R04和缺陷型R.R04细胞形成不完整隔膜的比例增加;造成96.4%的大肠杆菌BL21细胞表面凹陷,胞质内容物流失,菌体体积缩小;导致枯草芽孢杆菌89.6%的细胞体积明显缩小;导致金黄色葡萄球菌基本没有细胞能形成完整的分裂隔膜;导致红酵母细胞能进行出芽生殖的比例从64.2%降低到19.3%,但对其细胞形态无明显改变.联苯代谢物2,3-二羟基联苯对红球菌R04及其它微生物细胞分裂和增殖的抑制作用比其前体-联苯强,建议在研究环境化合物与微生物互作时,应考虑环境化合物代谢的毒性效应. 相似文献

Biodegradation and bio-sorption of antibiotics and non-steroidal anti-inflammatory drugs using immobilized cell process 总被引:2,自引:0,他引:2
In the present study, the removal mechanisms of four antibiotics (sulfamethoxazole, sulfadimethoxine, sulfamethazine, and trimethoprim) and four non-steroidal anti-inflammatory drugs (acetaminophen, ibuprofen, ketoprofen, and naproxen) in immobilized cell process were investigated using batch reactors. This work principally explores the individual or collective roles of biodegradation and bio-sorption as removal routes of the target pharmaceuticals and the results were validated by various experimental and analytical tools. Biodegradation and bio-sorption were found as dominant mechanisms for the drug removal, while volatilization and hydrolysis were negligible for all target pharmaceuticals. The target pharmaceuticals responded to the two observed removal mechanisms in different ways, typically: (1) strong biodegradability and bio-sorption by acetaminophen, (2) strong biodegradability and weak bio-sorption by sulfamethoxazole, sulfadimethoxine, ibuprofen and naproxen, (3) low biodegradability and weak bio-sorption by sulfamethazine and ketoprofen, and (4) low biodegradability and medium bio-sorption by trimethoprim. In the sorption/desorption experiment, acetaminophen, sulfamethoxazole and sulfadimethoxine were characterized by strong sorption and weak desorption. A phenomenon of moderate sorption and well desorption was observed for sulfamethazine, trimethoprim and naproxen. Both ibuprofen and ketoprofen were weakly sorbed and strongly desorbed. 相似文献

Identification of key parameters controlling dissolved oxygen migration and attenuation in fractured crystalline rocks 总被引:1,自引:0,他引:1
In the crystalline rocks of the Canadian Shield, geochemical conditions are currently reducing at depths of 500-1000 m. However, during future glacial periods, altered hydrologic conditions could potentially result in enhanced recharge of glacial melt water containing a relatively high concentration of dissolved oxygen (O2). It is therefore of interest to investigate the physical and geochemical processes, including naturally-occurring redox reactions, that may control O2 ingress. In this study, the reactive transport code MIN3P is used in combination with 2k factorial analyses to identify the most important parameters controlling oxygen migration and attenuation in fractured crystalline rocks. Scenarios considered are based on simplified conceptual models that include a single vertical fracture, or a fracture zone, contained within a rock matrix that extends from the ground surface to a depth of 500 m. Consistent with field observations, Fe(II)-bearing minerals are present in the fractures (i.e. chlorite) and the rock matrix (biotite and small quantities of pyrite). For the parameter ranges investigated, results indicate that for the single fracture case, the most influential factors controlling dissolved O2 ingress are flow velocity in the fracture, fracture aperture, and the biotite reaction rate in the rock matrix. The most important parameters for the fracture zone simulations are flow velocity in the individual fractures, pO2 in the recharge water, biotite reaction rate, and to a lesser degree the abundance and reactivity of chlorite in the fracture zone, and the fracture zone width. These parameters should therefore receive increased consideration during site characterization, and in the formulation of site-specific models intended to predict O2 behavior in crystalline rocks. 相似文献

The biokinetics of attached and suspended bacteria are an essential component of activated sludge models, anaerobic digestion models and biofilm models. These parameters are often assumed or “confirmed” based on the goodness-of-fit of the bioprocess models. Using a microbial fuel cell with a baffled reactor chamber, the attached- and mixed-growth microbial decay coefficients were evaluated under anaerobic conditions. The capability for real-time voltage recording allows easy and accurate measurement of the anaerobic microbial decay coefficients (bL, lysis-regrowth approach), which were determined to be 0.11 ± 0.01 and 0.15 ± 0.01 d−1 for attached (to anode) and mixed (present in the anode chamber) growth microorganisms, respectively. The corresponding half-saturation constants using glucose as a substrate were 204 ± 10 and 123 ± 1 mg COD l−1. Hence, like an oxygen uptake rate-based approach to measure the microbial kinetics under aerobic conditions, the electrochemical recording provides an attractive method to measure anaerobic microbial decay coefficients. 相似文献

Recent research and technology have provided promising outcomes to rely on biodiesel as the alternative and conventional source of fuel. The use of renewable sources constitutes the main stream of research. Waste Cooking Oil (WCO) was used for biodiesel production in this study. Lipase enzyme producing fungi Rhizopus oryzae 262 and commercially available pure lipase enzyme were used for comparative study in the production of FAAE. The whole cell biocatalyst and pure enzyme were immobilized using calcium alginate beads. It was prepared by optimizing with different molar ratios of calcium chloride and different percent sodium alginate. Entrapment immobilization was done for whole cell biocatalyst. PE was also immobilized by entrapment for the transesterification reaction. Four different solvents methanol, ethanol, n-propanol, n-butanol were used as the acyl acceptors. The reaction parameters like temperature, molar ratio, reaction time, and amount of enzyme to be used were also optimized for methanol alone. The same parameters were adopted for the other acyl acceptors too. Among the different acyl acceptors, methanol whose reaction parameters were optimized showed maximum conversion of triglycerides to FAAE - 94% with PE and 84% with WCB. On the whole, PE showed better catalytic converting ability with all the acyl acceptor compared to WCB, further study, it was observed that three consecutive and reversible reactions occurred in the interesterification of triglycerides. So, a kinetic model based on Michaelis-Menten equation with competitive substrate inhibition was used to find the maximum reaction rate Vi for the four solvents using pure enzyme and WCB. 相似文献

以剩余污泥为阳极底,糖蜜废水为基质,Ag Cl废水为阴极电子受体,构建了双室微生物燃料电池(Two-chamber Microbial Fuel Cell,简称MFCs),并研究了电池的产电特性、库仑效率及金属去除率.结果表明:Ag+不仅可以作为阴极电子受体,而且还能稳定产电,外电阻为1000Ω时,获得的最大电压为514.5 m V,最大功率密度为65.82 m W·m-2.在阴极实现了对废水中Ag+的去除,最大去除率可达71.6%,而且Ag+浓度为2000 mg·L-1时,回收金属银单质质量为197.66 g.在阳极对废水的处理效果也很显著,库仑效率最高为2.66%,COD去除率最大为81.22%. 相似文献

The molecular basis of male reproduction for cross-regulation between androgen and thyroid hormone axes is still rudimentary. This study aims to define a possible mechanism of hypothyroidism-induced reproductive influence with respect to sex hormone, mineral, sperm motility, oxidative stress, c-Fos expression, cell cycle, and apoptosis in rat testes. The Wistar rats were randomly divided into control group (NS) and hypothyroidism group [1 ml/100g BW/day, 0.1% propylthiouracil (PTU)] by intragastric gavage for 60 days. Blood samples were collected to measure the serum levels. The epididymis was excised to measure sperm motility and testes were excised to measure mineral, oxidative stress, c-Fos expression, cell cycle, and apoptosis. After 60 days, body weight, relative testes weight, triiodothyronine, and total thyroxine were all significantly decreased, whereas thyroid stimulating hormone was increased in the hypothyroidism group. A significant increase in sex hormone level of estradiol (E2) and significant decreases in testosterone (T) and T/E2 ratio were observed following PTU treatment. And sperm quality was also significantly changed. There were significant decreases in the contents of calcium (Ca2+) and zinc (Zn2+). On the other hand, malondialdehyde and hydrogen peroxide contents significantly increased, whereas the activity of superoxide dismutase, catalase and nitric oxide synthase and nitric oxide content significantly decreased in hypothyroid rats. The mRNA and protein expressions of c-Fos decreased significantly. The cell percentage in G0/G1 phase increased significantly, whereas decreased significantly in S and G2/M phases. Also, a significant increase in testicular cell apoptosis was observed in hypothyroid-treated rats. These results suggested that hypothyroidism could affect reproductive function in the form of changed sex hormone levels, sperm motility and testicular Ca2+ and Zn2+, and enhanced oxidative stress leading to c-Fos abnormal expression and increased apoptosis. 相似文献

本研究以天津泰达污水处理厂污泥浓缩间的污泥为接种物,启动并运行了微生物燃料电池(MFCs).从富集的阳极生物膜上分离得到了一株纯培养的微生物菌种,命名为P2-A-5.研究发现,菌株P2-A-5的16S rDNA序列与菌株Kocuria rhizophila DC2201具有100%的同源性,结合该菌的形态特征和生理生化实验,将其归属为嗜根考克氏菌(Kocuria rhizophila).通过化学剂处理、底物种类和浓度的优化,进一步提高其在微生物燃料电池中的产电性能.结果表明,菌株K.rhizophila P2-A-5经0.5 mg·L-1溶菌酶处理45 min后,接种到以2.0 g·L-1海藻糖为底物的阳极液中运行MFCs,其功率密度达到314.8 m W·m-2,比优化前(74.9 m W·m-2)提高了320.3%.这是首次对K.rhizophila种内微生物产电性能及其在微生物燃料电池中应用的报道,其成果对于丰富产电微生物的多样性,挖掘更多具有高电化学活性的微生物菌种,提高其产电性能具有重要的理论意义. 相似文献
